Given the serious risks of gonococcal disease, both sexually transmitted and obstetric, there is a need for a safe and effective vaccine to prevent gonorrhea. The approach proposed here is to develop a vaccine that can be used to produce a protein that would be targeted to the membrane of the gonococcus where it could function as a receptor and mediate gonococcal uptake into the host cell. The approach that will be used here is to develop a novel, multi-subunit vaccine targeting the gonococcal outer membrane. The outer membrane is the structural interface between the gonococcus and the environment, thus serving as the principal point of entry for gonococci into the host cell. The specific aims of this project are: 1. To use molecular biology techniques to construct three recombinant proteins that are antigens of the gonococcal outer membrane, namely porin-1, OM-associated protein (OmpA) and porin-2;2. To test the ability of the three recombinant proteins to promote antibody and T-cell responses in mice;and 3.
